Effect of Scalp Acupuncture on the Expression of IL-6 and IL-10 in Hippocampal CA1 Region of Acute MCAO Rats

Abstract: Objective: To detect the expression of scalp-acupuncture on the expression of IL-6 and IL-10 of hippocampal CA1 region in cerebral ischemia (CI) rats, so as to investigate its mechanism underlying regulation of Inflammation of ischemia stroke. Methods: 40 SD rats were randomly allocated to normal group, model group, scalp-acupuncture group and drug group(n=10 in each group). The model of focal CI was established by middle cerebral artery occlusion(MCAO). Scalp-acupuncture stimulation was applied to bilateral Dingnieqianxiexian (MS6) and Dingniehouxiexian (MS7) once daily for 7 days. Drug group were injection of pyrrolidine dithiocarbamate 100mg.kg-1/d, once daily for 7 days. The expression of IL-6 and IL-10 was detected by immunohistochemistry. Results: Compared with the normal group the expression of IL-6 and IL-10 in the hippocampal CA1 region significantly increased in the model group (P<0.05), while the expression of IL-6 in the hippocampal CA1 region obviousiy decreased in both scalp-acupuncture and drug group compared with the model group but the expression of IL-10 increased(P<0.05); Compared with drug group, the expression of IL-6 and IL-10 in hippocampal CA1 neurons of rats was not statistically significant(P>0.05). Conclusion: Scalp-acupuncture stimulation can increase the expression ofantiinflammatory factor IL-10, while decrease the expression of inflammatory factor IL-6 of ischemia stroke,which may be one of the molecular mechanisms of acupoint stimulation of acute ischemic cerebrovascular disease.
