The Effect of Moxibustion on Proliferation and Differentiation of Neural Stem Cellsin Vascular Dementia Rat Hippocampus

Abstract: Objective: To observe the effect of moxibustion at GV14, GV20 and GV24 on NSCs’ proliferation  and  differentiation  in  VD  rats’  hippocampus.  Methods:Establishing  vascular dementia  model  in  rats  by  ligating  right  and  left  common  carotid  artery  successively  after repeatedly ischemia-reperfusion.Cultivating and passaging neural stem cells and endothelial cells respectively,  marking  neural  stem  cells  with  lentivirus  mediated  EGFP,  and  constructing  cotransplanted implant in virto for celluar transplantation. 24 model rats were divided into 1 groups randomly as NE group, NEM group. And each group  was divided into 2 parts by the treatment intervention  period  as  T1  and  T2.  Rat  brains  were  taken  on  time  after  perfusion  of  4% paraformaldehyde.  Marker  protein  expression  was  detected  by  laser  confocal  microscope  to analyse the moxibustion effect of neurogenesis.  Results:The MAP expression in NE group was significantly decreased in T2 than in T1 (P<0.05). The T2 MAP expression in NEM group was significantly  higher  than  in  NE  group  (P < 0.05).  The  NeuN  expression  in  NE  group  was significantly decreased in T2 than in T1 (P<0.05). The T2 NeuN expression in NEM group was significantly higher than in NE group (P<0.05). Conclusion: Moxibustion can promote NSCs’ proliferation and differentiation in VD rats’ hippocampus.
