Effect of Electroacupuncture on Learning and Memory in Mice with High Glucose

Abstract: Objective: To observe the effect of Electroacupuncture on the behavior of learning and memory in diabetic mice, and to investigate the Protective effects of Electroacupuncture on the neurons in high glucose. Methods: A total of 48 8-week-old male C 57 BL/6 mice were randomly divided into sodium chloride group, sodium chloride + electroacupuncture group, diabetic model group (STZ), diabetic model Plus Electroacupuncture group, 12 rats in each group. Diabetes model was induced by 150  mg/kg streptozotocin (STZ) injection. Electroacupuncture at "Baihui" (GV 

20) "Shenshu"(BL 23)   "Zusanli"(ST 36),   voltage 2~4 V, frequency 2 Hz, once every other day, 20 min each time, 28 consecutive D. After 28 D, the mice were subjected to water maze and Passive avoidance test, and the density of dendrites of neurons in mice was observed by Golgi staining. Results: The diabetic model group compared with the sodium chloride group increased blood glucose (P<0.05) and insulin resistance (P<0.05), glucose (P<0.05) and insulin resistance (P<0.05),diabetic  model  +  EA  group  compared  with  the  diabetic  model  group  reduced  blood glucose  (P<0.05),  elevated  ins  ulin  levels  (P<0.05);  water  maze  test,28,  29  D  navigation experiment Part, the diabetic model group compared with the sodium chloride group escape the latency was increased (P<0.05), diabetic model + EA group compared with the diabetic model group  (P<0.05),  the  escape  latency  decreased  30,  31  D  of  space  exploration  experiment,  the diabetic model group compared with the sodium chloride group escape  latency increased (P<0.05), diabetes mellitus model + EA group compared with the diabetic model group (P<0.05), the escape latency decreased 32 D on space exploration of experimental diabetic model group compared with the  sodium  chloride  group  increased  escape  latency  (P<0.05),  diabetic  model  +  EA  group compared  with  the  diabetic  model  group  to  avoid  Potential  Ummer  reduced  (P<0.05),  in  the Passive avoidance test, during the 14 and 28 d memory test, the diabetic model group compared with the sodium chloride group was reduced (P<0.05), diabetic model + EA group compared with the diabetic model group increased latency (P<0.05); diabetes model group compared with the sodium chloride group reduction of dendritic spine density (P<0.05), model + EA group compared

with  the  diabetic  model  group  increased  dendritic  density  (P<0.05).Conclusion Electroacupuncture  can  improve  diabetic  cognitive  dysfunction,  which  may  be  related  to regulating the density of neuronal dendrites.
