The Effect of Wrist-ankle Acupuncture on the Expression of 5-HT3A in Spinal

Abstract: Objective: To observe the effect of wrist-ankle acupuncture and electroacupuncture on pain and pain behavior of rats in bone cancer and effects on 5-HT3A in spinal cord area.  Methods:  The 32 SPF female Wistar rats were randomly divided into four groups: electroacupuncture group(EA), wrist-ankle acupuncture group(WA), cancer pain group(CA) and sham operation group(Sham). The bone cancer pain model was established in the left leg of the electroacupuncture group, the wrist-ankle acupuncture group and the cancer pain group. The sham operation group was injected with the inactivated tumor cells in the left leg tibial intramedullary cavity.  The  electroacupuncture  group  and  the  wrist-ankle  acupuncture  group  began  the experimental intervention on the 9th day after the molding. No intervention after operation of cancer pain group and sham operation group. The electroacupuncture group was treated with the left "post three mile" and  "heel"  point,  which  was  unclogging,  2Hz,  and  intensity  2  daily,  each  time  for  30  min,  continuous treatment  for  6  days. Wrist-ankle  needle  group,  take  left  "heel"  point,  the  needle  from  4  PM  to  8  PM, continuous treatment 6 days. All the rats were weighed on the next day. The mechanical pain threshold was determined one day before surgery, 9 days after surgery, 11 days, 13 days and 15 days. He staining was used to detect the changes of tibial histology in rats. The expression of 5-HT3A mRNA in rat spinal cord was detected by RT -PCR. The expression of 5-HT3A receptor protein in spinal cord was detected by Western blot method. Immunohistochemistry was used to observe the expression of 5-HT3A in the region of the spinal cord. Results: Weight: after molding, the body weight dropped on the 6th day after the formation of the electroacupuncture group, wrist-ankle acupuncture group and cancer pain group, and the group weight of the sham group showed a trend of growth. In the 10th and 12th days, electric acupuncture needle, wristankle acupuncture and pain group no difference between the three groups of weight, the 14th day electric acupuncture  group  and  wrist-ankle  acupuncture  pain  needle  group  and  compared  with  a  significant difference (P<0.01), the curative group and wrist-ankle acupuncture group rats weight rebound, but the pain group weight did not see obvious rise trend. Mechanical pain: no difference in basal pain (P>0.05). In the 9th day, the pain was significantly different ( P<0.01), the sham operation group was superior to the other three groups, and there was no significant difference between the electroacupuncture group and the wristankle acupuncture group and the cancer pain group (P<0.05). On the 11th day, the wrist-ankle acupuncture group was better than the electroacupuncture group and the cancer pain group. On the 13th day, the wristankle acupuncture score was superior to the electroacupuncture group and the cancer pain group. On the 15th day, the score of the electroacupuncture group and the wrist-ankle acupuncture was superior to the cancer pain group (P<0.01). The non-surgical side was superior to  the electroacupuncture group and the cancer  pain  group on  the 15th  day.  RT -PCR  results:  16  days,  spinal  cord  dorsal horn  5-HT3A  mRNA expression  there  are  differences  between  the  four  groups  (P<0.01),  the  curative  group  and  wrist-ankle acupuncture group  had significant difference compared with pain group (P<0.05), the curative group and wrist-ankle acupuncture not obvious difference (P>0.05). Results of WB: the expression of 5-HT3A receptor protein in the 16th day of the spinal cord was significantly different in the fourth group (P<0.01), and the sham operation group was significantly lower than the other three groups. Both the electroacupuncture group and  the  wrist-ankle  acupuncture  group  were  lower  than  the  cancer  pain  group  (P<0.01).  Moreover,  the expression of 5-HT3A receptor in the spinal cord was observed, and there was significant difference between the  electroacupuncture  group  and  the  wrist-ankle  acupuncture  group  compared  with  the  sham  operation group (P<0.01). Immunohistochemistry results: in the 16th day, the 5-HT3A receptor was observed mainly in the superficial layers of the sensory stimulation. Dorsal horn of spinal cord area positive area statistics, found that the curative group, wrist-ankle acupuncture pain and spinal cord dorsal horn region positive rate were higher than that of control group (P<0.05), and electric acupuncture group and wrist-ankle acupuncture group and compared with the positive rate in difference (P<0.05). The electroacupuncture group and the 

wrist-ankle  acupuncture  group  were  not  obvious.  Conclusion:  The  electroacupuncture  and  wrist-ankle acupuncture improved the mechanical pain threshold of the bone cancer pain rats, and improved the weight status of the model rats. The mechanism may be related to the expression of 5-HT3A in the shallow region of the spinal cord with the electroacupuncture and wrist-ankle acupuncture.
