The Study of Acupuncture Preconditioning on the Opening Rgulation Mechanism of Mitochondrial Permeability Transition Pore in Rat Ischemic Myocardium

Abstract: Objective: To investigate the molecular mechanism by study of Xinshu electrical acupuncture preconditioning on the open regulation mechanism of mitochondrial permeability transition pore in rat ischemic myocardium . Methods: Rats divided into blank group, model group, xinshu acupuncture preconditioning group , ischemic preconditioning group by random number table corresponding. Duplicate myocardial ischemia and reperfusion injury model by alive rat heart ligation of left anterior descending coronary artery . Blank group: Rats are fixed with acupuncture preconditioning group for 30 minutes but not acupuncture before molding;once per day,continue 15 days. Model group: Rats are fixed with blank group before molding; No other factors were imposed on the whole experiment except myocardial ischemia reperfusion.Acupuncture preconditioning group: Acupuncture 15 days until molding; once per day, 30 min every time,then MIRI molding.Ischemic preconditioning group: Dealing with rats the same as the blank group before molding; To block blood flow 5 min then perfusion 5 min before coronary artery ligation on molding. Repeated 3 times in a row, a total of 30 min ischemic preconditioning. Detect myocardial cell mitochondrial membrane permeability transition pore opening degree after myocardial ischemia molding success 0 h, 24 h, 48 h. Results: 1. The model group dyed myocardial mitochondria green fluorescent compared with the blank group was reduced obviously by laser confocal microscope observing, the experiment result shows that the mitochondrial membrane permeability transition pore activity of model group was significantly reduced (P < 0.01). 2. Ischemic preconditioning 0h group and acupuncture preconditioning 0h group mitochondrial fluorescence intensity is significantly reduced compared with the model group by laser confocal microscope observing, but the indicators have no significant difference between ischemic preconditioning 0h group and acupuncture preconditioning 0h group (P> 0.05).3. Acupuncture preconditioning 24h, 48h group mitochondria fluorescence intensity have a certain enhancement (P < 0.05 or P<0.01) observed by laser confocal microscope compared with ischemic preconditioning group and acupuncture preconditioning 0h group.4. Experiment data shows that acupuncture preconditioning group and ischemic preconditioning group both can reduce myocardial ischemia reperfusion injury; And perhaps acupuncture preconditioning 24 h group is the best treatment plan. Conclusion: 1. The protection of electrical acupuncture Xinshu preconditioning for myocardial ischemia after 0h,24h,48h was different, the therapy effect enhance - peak - attenuated trend increased with time,and perhaps acupuncture preconditioning 24 h group is the best treatment plan.2.Electrical acupuncture Xinshu has protective effect for MIRI ,the mechanism may be by inhibiting mPTP opening in myocardial ischemia reperfusion injury and reducing the mitochondrial apoptosis then reduce the myocardial ischemia injury.
