Effects of Electroacupuncture Pretreatment on mRNA Expression of TLR4、MyD88 and NF-кB in the PC6 Area of Rats with Myocardial Ischemia-reperfusion Injury

Abstract: Objective:To observe the effect of electroacupuncture(EA) pretreatment on mRNA expression of TLR4、MyD88 and NF-кB of “Neiguan” Point(PC 6)in the PC 6 area of rats with myocardial ischemia-reperfusion injury. Methods : The healthy wistar rats were randomly divided into 6 groups, i.e., the control group, the normal EA group, the sham operation group ,the model group, the EA group, the sham EA group, 8 in each group. The rats in the EA group, the normal EA group and the sham EA group were treated with EA (2~20 Hz in frequency, tolerated intensity of rats) at “Neiguan” (PC 6), 30 min per treatment, once a day for consecutive 5 days. The rats in the sham operation group and model group were treated with immobilization for the same time, and no EA was given. Animals were subjected to 30 min of ischemia induced by occluding the left anterior descending artery followed by 120 min of reperfusion after the end of EA. The mRNA expression of TLR4, MyD88 and NF-κB in the PC6 area were determined with Real-time PCR. Results : Compared with the normal group, the ECG J-point height were significantly increased in the model group (P<0.01), compared with the model group and the sham EA group ,EA preconditioning can significantly reduced ECG - J point height(P<0.01); The mRNA expression of TLR4, MyD88 and NF-κB were significantly increased in the model group and the sham EA group compared with the control group (P<0.01). EA preconditioning significantly reduced the the mRNA expression of TLR4, MyD88 and NF-κB compared with the model group and the sham EA group (P<0.05). Conclusion: EA preconditioning significantly reduced the the mRNA expression of TLR4, MyD88 and NF-κB in the PC6 area of rats with myocardial ischemiareperfusion injury.
